CL rotor change (gts) question

So I was looking into changing rotors on my gts... Question- the red nipples that the wheel goes on... Do they need to come off ? If so, any special instructions or things to do here? This is one more step required for CL...

The manual just says that all contact surfaces must be clean and free of grease, then tighten the self locking screws to 6Nm.
