weight first and foremost ... and (in theory at least) rotational inertia ... probably too small to detect with the "seat of the pants" tricorder ...
comfort level, in order to maintain rolling circumference, you will need a lower profile tire on a 20" vs. a "19, which means that the effective spring rate of the 20" will be higher (aka stiffer/firmer) again probably not entirely noticeable...
